The foundation of Guayaquil was a process of Spanish conquest in the current coastal region of Ecuador, which, after several transfers and conflicts with native inhabitants of pre-Columbian cultures, it was finally founded the city of Santiago de Guayaquil. The conquest took place between 1534 and 1547, during which it was changing its name according to the place of establishment, retaining the name of their patron saint.

The founding process occurred after the entry of Diego Teran to the northern mountains of old Tahuantinsuyo Inca, sent by the Marquis Francisco Pizarro from Cuzco, with the aim of accelerating the settlement of these territories and fortify to counter possible invasion of advance Pedro de Alvarado, who came from Central America. Almagro founded the city of Santiago de Quito on August 15, 1534, near the present city of Riobamba in Ecuador. Following negotiations between Almagro and Alvarado, Santiago had to be moved to another site because it did not meet the requirements imposed by the King of Spain in the Royal Decree issued on May 4 of that year, thereby appointed the departure of two expeditions.

The first expedition took northwards and founded the town of San Francisco de Quito on December 6 of that year by Sebastian Benalcázar. In mid-1535, he left the other expedition bound for the coast, however, took southbound back to Piura. Benalcázar brought together several men and supplies by sea to sail the Gulf of Guayaquil and the Guayas River traced to move the city of Santiago in the region.

The natives of the region were the main obstacle to the establishment of the city, because they resisted several times in the presence of the Spaniards. Benalcázar population founded by late 1535 was destroyed by fire caused by chongos; then its reconstruction and transfer into the hands of Hernando de Zaera in 1536 was also attacked on many occasions, leaving heavy casualties among the population; later, in 1542 Captain Francisco de Orellana ordered and carried out the transfer of the city to another sector, however, after his departure took with him several soldiers leaving the population unprotected, so Santiago Diego de Urbina moved again to another location. Huancavilcas ravaged the residents, who were forced in 1543 to move closer to the first settlement of Benalcázar again. Finally, due to the civil wars between Spaniards finally it decided to move it in its current location to the west bank of the Guayas River.
